CD40蛋白是一种属于肿瘤坏死因子受体(TNF-R)超家族的跨膜糖蛋白,广泛表达于B细胞、树突状细胞、单核/巨噬细胞、某些上皮细胞及多种肿瘤细胞表面,其配体为CD40L(CD154),主要由活化的CD4⁺ T细胞表达。CD40–CD40L相互作用在体液免疫和细胞免疫中起关键作用,可促进B细胞活化、抗体类别转换、生发中心形成以及记忆B细胞的产生,同时增强抗原提呈细胞的共刺激能力和促炎因子分泌,从而放大免疫反应。CD40信号通路可激活NF-κB、MAPK等多条下游通路,参与炎症反应、自身免疫疾病发生以及肿瘤免疫调控,因此成为疫苗佐剂、免疫治疗和抗肿瘤药物研发的重要靶点。
