GLP-1受体(GLP-1R)是一种位于胰腺细胞和大脑神经元上的G蛋白偶联受体,由6号染色体上的GLP1R基因编码,属于胰高血糖素受体家族。激活GLP-1R会刺激腺苷酸环化酶(AC)通路,增加细胞内cAMP水平,激活蛋白激酶A(PKA),从而促进胰岛素的合成与释放,因此GLP-1R是糖尿病治疗药物(GLP-1R激动剂)的重要靶标。此外,GLP-1R也参与大脑中的食欲调控。

当GPCR被激动剂激活后,其胞内C端尾部及特定磷酸化环会被G蛋白偶联受体(GRK)磷酸化,改变受体构象并暴露出与β-arrestin结合的位点。Β-arrestin在静息状态下处于自抑制构象,磷酸化后其C端尾部从N端释放,暴露出结合位点。Β-arrestin通过其C端结构域自发结合到细胞膜的脂质双分子层,并通过横向扩散与膜上的GPCR短暂相互作用,随后在配体激活下,β-arrestin与受体在细胞膜上的驻留时间显著延长,促进其向网格蛋白包被小窝(CCPs)的共定位,最终触发受体内吞。这一过程不仅介导GPCR脱敏和内化,还可通过支架功能激活非G蛋白依赖的信号通路。
