降钙素基因相关肽家族,包括降钙素(CT, Calcitonin)、胰岛淀粉样肽(AMY, Amylin)、降钙素基因相关肽(CGRP, Calcitonin Gene Related Peptide) 、肾上腺髓质素(AM, Adrenomedullin) 。
Amylin有三类潜在受体,均是由降钙素受体(CALCR、CTR)和受体活性修饰蛋白(RAMPs)形成的复合物。受体活性修饰蛋白(RAMPs)是一系列I型单跨膜蛋白,它们与G蛋白偶联受体形成异源二聚体,分为三种亚型:RAMP1,RAMP2和RAMP3。它们具有较大的细胞外氨基末端结构域,单个TM跨膜结构域和短的〜10个氨基酸的胞质结构域。这三个RAMP具有相似的基本结构,但氨基酸序列中只有约30%的同一性。CALCR和RAMP1形成的复合物AMY1, CALCR和RAMP2形成的复合物AMY2, CALCR和RAMP3形成的复合物AMY3。
Amylin由胰腺产生,并作为激素发挥作用,调节营养摄入。 Amylin神经元沉积的研究主要集中在Amylin在阿尔茨海默病中的作用。
Amylin与受体复合物结合后,复合物与G蛋白相互作用,Gαs亚基从复合物中分离并激活腺苷酸环化酶,产生cAMP。 细胞内cAMP浓度的升高刺激cAMP依赖的信号通路,最终导致转录因子cAMP反应元件结合蛋白与启动子结合,诱导基因表达。
