THRβ(Thyroid Hormone Receptor Beta,甲状腺激素受体β)是一种配体依赖性的核受体转录因子,属于核激素受体超家族成员。THRB基因在人类中位于染色体3p24.2,其产物主要作为甲状腺激素T3(三碘甲腺原氨酸)的受体。THRB蛋白通过与T3结合,调控下游靶基因的转录,从而影响多种生理过程,包括代谢调控、发育、心血管功能以及神经系统的成熟。THRB可以与视黄酸X受体(RXR)形成异源二聚体,结合到特定的DNA反应元件上,实现激素信号的精确调控。

THRβ具有多个剪接变体,主要包括TRβ1和TRβ2,两者在组织分布和功能上有所不同:TRβ1主要存在于肝脏、肾脏和心脏等外周组织,TRβ2则主要分布于下丘脑、垂体和视网膜。THRB基因突变与甲状腺激素抵抗综合征、代谢异常及多种肿瘤的发生相关,被视为潜在的代谢与癌症治疗靶点。
