| Background | The CD8 gene, located in the 2p12 region of the human genome, encodes a CD8 transmembrane glycoprotein (belonging to the immunoglobulin superfamily) consisting of α/β chains forming a heterodimer (CD8 αβ) or α chain homodimer (CD8 αα) with a molecular weight of approximately 32-37 kda. Its extracellular v-like domain specifically binds to the mhc-class I α3 domain, which is involved in T cell activation signal transduction as a TCR co-receptor, and the intracellular segment initiates calcium ion flow through the phosphorylation of ITAM motif by P56lck kinase, which is involved in the activation of T cells, drives CD8 + T cell differentiation into cytotoxic T lymphocytes (ctls) . Functionally, CD8 + T cells play a central role in antiviral and antitumor immunity by releasing perforin/granzyme to directly kill virally infected cells or tumor cells, and secreting cytokines such as IFN-γ to regulate immune responses, which may be related to the role of CD8 + T cells in anti-viral and anti-tumor immunity, it is also involved in the mechanisms of autoimmune diseases (such as rheumatoid arthritis) , transplant rejection and tumor immune escape. |
