FGFR2b(成纤维细胞生长因子受体2b)是成纤维细胞生长因子受体家族(FGFR)的重要成员,由FGFR2基因通过选择性剪接产生的上皮特异性异构体,其胞外结构域包含三个免疫球蛋白样结构域(D1-D3),特异性结合FGF7、FGF10等配体,通过激活下游RAS-MAPK、PI3K-AKT/mTOR及STAT3等信号通路调控细胞增殖、分化、迁移及血管生成。该受体主要在上皮细胞中表达,与间质来源的FGFR2c(IIIc剪接变体)形成旁分泌信号轴,参与胚胎发育、器官形成及组织修复等生理过程。病理状态下,FGFR2b的异常激活(如基因扩增、突变或配体过表达)可驱动肿瘤发生发展,约30%-38%的胃癌、乳腺癌、胆管癌等上皮源性肿瘤存在FGFR2b过表达,且与侵袭性表型、淋巴结转移及不良预后显著相关。
